1 Game? I couldn't decide, erm, here's a short list of the highlights I'm looking forward to:

Supreme Commander
Super Mario Galaxy
Metroid Prime 3
Super Smash Brothers Brawl
Resident Evil (Umbrella Chronicles and 5)
Time Splitters 4
